ネコ免疫不全FIVの抗酸化剤治療の基礎的検討と臨床応用

研究概要	本研究はヒトエイズのネコのホモログであるFIVにおいて、FIVのウイルス増殖の際生じる活性酸素の上昇に呼応して活性化するNFκBなどの転写因子にレドックス制御が関与するかどうか、抗酸化物質で抑制されるのか、さらには、それが、実際の疾患に対して、抗酸化物質が治療効果を増強できるのかを明らかにする目的で実験を行った。初年度は酸化ストレスによって転写因子NFκBやAP-1などの活性化のアッセイ系を確立し、抗酸化剤の効果をインビトロで確立することを目的とした。モデル系として牛血管内皮細胞とMOLT-4細胞を用いて、過酸化水素処理あるいは放射線照射による転写因子NFκ-BとAP1の活性活性化のゲルシフトアッセイ法をもちいて検討した。NFκ-BならびにAP-1のDNA上流のコンセンサス領域の配列の20mer程度のDNAを用意し、T4キナーゼで^<32>P末端ラベルし、細胞の核抽出物と一緒にインキュベーションし、中性条件下で電気泳動を行い、NFκ-BがDNAと結合するとゲル上で移動度が遅くなる(シフト)ことでその活性を評価する方法である。その結果、過酸化水素1mM処理あるいは放射線照射でスーパーシフトしたバンドがそれぞれ観察され、抗NFκB抗体あるいは抗c-Jun抗体処理で消失した。また、別の転写因子活性の測定法として分泌型アルカリフォスファターゼの遺伝子の上流にNFκB,AP-1,NF-AT,CRE,MYC,GREあるいはSREなどの転写因子のコンセンサス配列を組み込んだレポーターアッセイシステムを導入し、測定系を確立した。また、これら転写因子活性化に対する抗酸化剤の効果ではαフェニルブチルニトロン(PBN)が酸化ストレス誘導AP-1の活性化を抑制することが明かとなった。次年度はネコ白血球を用いて、研究を発展させて行く予定である。
