血液―脳バリヤ―としての酸性スフィンゴ糖脂質の機能の解析

研究概要	1)第一に、GM2/GD2合成酵素遺伝子KOマウスとGD3合成酵素遺伝子KOマウスを交配して、ダブルKOマウスを作成し、特に外見に異常なく出生し成長することが確認された。 2)脳、肝臓、皮膚などから糖脂質を抽出し、その組成変化を検討したところ、ダブルノックアウトマウスではいずれの組成の酸性画分でもGM3のみが検出され、想定された合成経路に一致したガングリオシド組成の変化が確認された。 3)ダブルノックアウトマウスのニューロン/グリア相互反応を解析するために、脊髄後根神経節の細胞の初代培養法を検討した。Ara-Cを添加しない培養系ではグリア細胞の著明な増殖が得られ、GFAP染色で陽性を確認した。またAra-CとNGF存在下で培養した場合は、ニューロンが突起伸長を伴って培養可能なことを確認した。今後ダブルKOマウス由来の細胞で検討を行うと共に、野生型マウス由来のグリア細胞とダブルノックアウトマウス由来のグリア細胞におけるニューロン支持機能の差異につき明らかにする。 4)更に、ダブルノックアウトマウスの顔面において難治性の皮膚損傷が見られたので、そのメカニズムの解明を行った。皮膚の切片標本で、異常な神経線維の増生が見られ、また、三叉神経、坐骨神経の膨化、ダンゴ状変化、変性死、不正形変化等が20週齢以前より認められた。これらの変化は、末梢神経におけるニューロンとグリア細胞あるいはシュワン細胞との相互反応の障害に基づく神経組織維持機構の破綻によることが示唆された。 5)ダブルノックアウトマウスにおける血管膜のバリヤー機能について、HRP法入法を中心に比較検討中である。
