成体マウス体細胞における血管前駆/幹細胞の探索と血管再生への応用

研究概要	我々は最近、マウスES細胞を用いて血管構成細胞の分化誘導と血管の再構成を試み、VEGF(vascular endothelial growth factor)の受容体の一つであるFlk1を発現する細胞を分化誘導、純化・再培養することにより、このFlk1陽性細胞が血管の構成細胞である内皮細胞および壁細胞双方の前駆細胞(血管前駆細胞)として機能し、同前駆細胞を用いてin vitroおよびin vivoにおいて、血管構造を形成できることを明らかにした。一方、近年造血幹細胞のみでなく、神経幹細胞、間葉細胞幹細胞、骨格筋幹細胞など様々な臓器レベルの幹細胞が、成体の体細胞内に存在することが発見された。本研究は、こうしたES細胞における我々の知見を元にし、新しく成体において同様の血管前駆細胞の存在を探索し、同細胞の同定と同細胞を用いた血管再生の可能性を探ることを目的としている。本研究課題において以下の研究成果を得た。ヒト臍帯血および乳房脂肪組織においてFlk-1陽性細胞を約0.1〜1%の濃度で検出した。この細胞群はCD34^-、c-kit^-、AC133^-、VE Cadherin^-であった。この細胞をFACSセルソーターにより純化回収し、コラーゲンIV、ラミニンあるいはフィブロネクチンコートディッシュ上で培養を続けた。その結果、VEGF存在下で培養を行うと細胞は敷石状の配列となりVE Cadherin^+の内皮細胞の出現が確認された。一方、血管平滑筋マーカーであるアルファ平滑筋アクチン(SMA)陽性の細胞は極めて少数であった。
