研究概要

(1)恒常的にαBクリスタリンとHsp27を高いレベルで発現しているU373 MGヒトグリオーマ細胞をプロテアゾーム阻害剤であるMG-132に曝露すると、αBクリスタリンとHsp27の合成が誘導されると同時に両シャペロンの分子内セリン残基のリン酸化ご高進し、おもに不溶性分画に蓄積した。新たにHsp27の三カ所のリン酸化部位(Ser-5,Ser-78,Ser-82)を認識する抗体を作製し、ウェスタンブロット法で解析すると、三カ所のセリン残基すべてのリン酸化が高進していた。形態学的には、MG-132処理によって、核周辺部に封入体(アグリゾーム)が形成され、その構成成分としてリン酸化型を含む両シャペロン、ユビキチン、プロテアゾーム、βチュブリンなどがビメンチンに囲まれた状態で検出された。
(2)両シャペロンのリン酸化はp38MAPKの阻害剤で阻止されることから、p38MAPKの下流にあるMAPKAPK2/3によってリン酸化されることが推測された、事実、このカスケードが活性化されていることが観察された。
(3)MG-132による両シャペロンの誘導およびリン酸化の促進と不溶性分画への蓄積およびアグリゾームの形成とは、リン酸化阻害剤を共存させても認められた。しかし、アグリゾームの形成は微小管脱重合促進剤であるノコダゾールを共存させると阻止された。すなわち不溶性成分として細胞質内に蓄積した未処理蛋白質は、微小管を介して核周辺部に集めら、アグリゾームを形成することが示唆された。しかし、両シャペロンのリン酸化の意義は不明である。
(4)現在リン酸化αBクリスタリンと相互作用する分子をtwo-hybrid法にて拾い出し、同定中である
