p53C末端ペプチドによる放射線・温熱癌治療増感の基礎的研究

研究概要

【目的】p53C末端ペプチドは変異型p53蛋白質のDNA(p53CON)結合能を回復させるとの報告がある。今回、変異型p53遺伝子保有癌細胞にX線あるいは温熱誘導アポトーシスを増強させることを目指し、合成p53C末端ペプチドと放射線/温熱治療を併用した新しい癌治療法の基礎的検討を行った。【方法】p53の表現型が正常型の舌扁平上皮癌細胞SASとp53欠損ヒト肺癌細胞H1299に変異型p53遺伝子(codon248,Arg→Trp)およびneoコントロールベクターを導入し実験に用いた。X線あるいは温熱処理する前に、二種の合成p53C末端ペプチド(アミノ酸残基361-382と353-374)をこれら細胞に導入し、アポトーシス誘導を調べた。アポトーシスの解析はヘキスト染色法およびアポトーシス関連蛋白質の免疫細胞染色法で行った。また、未処理あるいはX線/温熱処理した細胞から抽出した蛋白質にp53C末端ペプチド加えたin vitro gel shift assayを行った。【結果】正常型p53癌細胞は変異型p53癌細胞に比べX線あるいは温熱誘導アポトーシスの出現率が低いが、合成p53C末端ペプチドを併用すると、変異型p53癌細胞のX線あるいは温熱誘導アポトーシスが増強されることを見出した。また、合成p53C末端ペプチドを前処理すると、変異型p53癌細胞にもX線/温熱で誘導される活性型Caspase-3陽性細胞、断片化PARP陽性細胞の増加及びアポトーシスの増強が見られるようになった。p53C末端ペプチドは変異型p53蛋白質のDNA(p53CON)結合能を回復させるが、細胞にX線あるいは温熱処理するとその結合能はより増強されることが分かった。このような効果はp53N末端ペプチド(14-27)には見られなかった。【考察】p53C末端ペプチドによる変異型p53のDNA結合能の回復は放射線/温熱によるp53活性化で増強されることが示唆された。その結果、p53依存的なCaspase-3活性化を介したX線あるいは温熱誘導アポトーシスが増強されるものと考えられる。【結論】変異型p53癌細胞に対しp53C末端ペプチドを併用した放射線/温熱治療が有効であることが示唆された。
