CBPによる脂肪細胞分化・肥大化及びインスリン感受性の調節メカニズム

研究概要	我々は阻害剤(J. Clin. Invest. 108 : 1001-1013, 2001)及び欠損マウス(J. Biol. Chem. 276 : 41245-41254, 2001)を用いた解析から、高脂肪食はPPARγを介して脂肪細胞の肥大化、インスリン抵抗性を引き起こすことを示した。本研究では、PPARγの主要なcoactivatorであるCBP欠損マウスを解析し、脂肪蓄積やインスリン感受性における生理的な役割を検討した。CBPヘテロ欠損マウスは野生型マウスの2/3程度の体重を示した。本マウスの体重当たりの組織重量は白色脂肪組織においてのみ著減していたが、これは細胞数の減少ではなく、細胞のサイズが小さいことに起因していた。本マウスでは普通食、高脂肪食下共に、より良好な耐糖能、インスリン感受性を示した。これらは、レプチン感受性が充進し、インスリン感受性ホルモンであるアディポネクチン(Nature Medicine 7 : 941-946, 2001)の発現量、血中濃度が増加していたこと、及びインスリン抵抗性を惹起する遊離脂肪酸の血中濃度、TNFαの発現量が低下していたことによるものと考えられた。白色脂肪組織では脂肪酸流入に関わる分子の発現低下、肝臓・骨格筋・脂肪組織では脂肪合成酵素の発現低下及び脂肪酸酸化酵素やUCP2の発現増加を認め、これらが骨格筋、肝臓での中性脂肪含量の低下をもたらし、良好な耐糖能、インスリン感受性に寄与していると考えられた。以上よりCBPヘテロ欠損マウスの表現型は、PPARγなどを介した脂肪蓄積経路の抑制と、レプチンとアディポネクチン情報伝達経路の活性化を介した脂肪酸の燃焼とエネルギー消費の亢進によるものと考えられた(Nature Genetics 30 : 221-226, 2002)。
