研究概要	<柳田研> コンデンシンサブユニット変異体cnd2の表現型解析から、コンデンシン複合体がM期凝縮反応のみならず、間期機能、特にDNA障害修復、Cds1/Chk2活性化に必須であり、さらに間期コンデンシン機能の欠損はS期チェックポイントを活性化することを発見した。コヒーシンサブユニット、およびそのリクルートメント因子の温度感受性株では、姉妹染色分体のコヒージョン異常がキネトコアースピンドル相互作用の欠損をもたらし、その結果スピンドルチェックポイントの活性化が起こることを明確に示した。ヒト動原体たんぱく質hMis12を発見し、これが染色体の均等分配に必須な役割をはたし、CENP-Aとは独立に動原体クロマチンに取り込まれることをRNAi法で示した。また動原体タンパクのCENP-Aの動原体局在に細胞周期で制御される新規GATA因子が必須であることも示した。 <武田研> 相同組み換えや多くのDNA修復経路は、DNA合成を伴う。ヒトDNAポリメラーゼは、複製に関与するものも含めて少なくとも13種類存在する。どのポリメラーゼが、どの修復・組み換え経路に関与するかは、動物細胞では全く不明である。我々は、高頻度に標的組み換えが起こるニワトリBリンパ細胞株、DT40からpol zetaの欠損株を作製した。これは、動物細胞から初めて樹立されたpol zeta欠損株である。そして、pol zetaが、既に解明された損傷乗り越えDNA合成の役割のみならず、相同組み換えにも関与することを解明した。さらに、pol zetaは、増殖細胞において染色体DNAの構造維持に重要な働きを持つことも解明した。 <竹安研> 塩透析法によるクロマチン再構成技術を用いて、数kbから100kbまでのプラスミドDNAあるいは線状にしたDNA上にクロマチンファイバーを形成させて、原子間力顕微鏡により可視化した。その画像の解析から、プラスミドDNAの持つ負のスーパーコイルがクロマチン形成を促進することを見出した。また、大腸菌とヒト培養細胞をガラス基板上で処理してゲノム構造を露出させ、原子間力顕微鏡により解析することで、原核生物と真核生物のゲノム構造に共通する幅80nmの基本構造を検出した。大腸菌ゲノムの解析においては、細胞増殖期と休止期との間でのゲノム構造の変化を明らかにし、休止期のゲノムは高度に凝集している様子が観察された。 <西田研> 低分子量GTPasc Ranとその制御因子RCC1およびRan GAPが、タンパク質の核細胞質間輸送の制御に関わるだけではなく、染色体のpositioning、分裂装置形成、さらには核膜形成に必須の役割を担っていることを明らかにした。Plk1が、MPFの活性化因子であるCdc25CをM期前期においてリン酸化することにより、Cdc25Cの細胞質から核への移行を引き起こすことを制御していることを示した。Plk1がbipartite nuclear localization signal(NLS)を有しており、そのNLSにより核内存在が規定されていること、またPlk1の核内存在が細胞周期のG2期からM期への進行を制御していることを示した。 <石川研> 1.ヒトコヘーシン複合体の構成要素Rad21/Scc1の機能を明らかにするために、そのN末端欠損変異体がドミナントネガティブ体として機能することを示し、そのような変異体を発現している細胞では姉妹染色分体合着が行われないために、スピンドルチェックポイントが活性化されてメタ期停止がおこることを示した。 2.DNA末端結合反応に関わることが知られているKu蛋白質について、分裂酵母Ku80をコードする遺伝子pku80を単離し、Pku80蛋白質がテロメアに局在するためにはテロメアDNAの物理的末端が必要であることを示した。 3.テロメア短小化を含む様々な原因によってもたらされる細胞老化現象に、ストレスMAPKの一つのp38が共通して重要な役割を果たしていることを明らかにした。 <松本研> スピンドルチェックポイントは、紡錘糸(スピンドル)と染色体の接続の不備を検出し、細胞周期の進行を抑止する監視機構である。このチェックポイントの中心機能因子、Mad2、に注目し、この蛋白質が紡錘糸未接続の染色体動原体に局在すること、またこの局在にはMAD2-結合蛋白質、Mad1、が必要であることを示した。またMad2に直接結合する新規のヒト蛋白質、Cmt2、について機能解析を行い、Cmt2は、スピンドル接続完了にともないスピンドルチェックポイントを解除し細胞周期の進行を促進するという、このチェックポイントの制御機構について新しい概念を打ち出した。これらの結果はスピンドルチェックポイントのシグナル経路を理解するために不可欠な貢献である。
