脳における「匂い地図」と「嗅覚記憶」の解析

研究概要	(1)ラット嗅球の「匂い分子受容体地図」の解明ウレタン麻酔下のラットの嗅球において、内因性信号の光学的測定法を用いて、各種の匂い分子がどの糸球を活性化するかを調べた。現在までに分子構造が系統的に異なった約200種類以上の匂い分子を用いた。新しく開発した手術法を用い、嗅球の背側表面から外側表面にかけて広く露出し、嗅球外側地図の約60%の領域の探索を終了した。この結果(1)個々の糸球は、分子構造の類似した一群の匂い分子により活性化されること(2)匂い分子応答特性が似ている糸球群は、嗅球外側地図の特定の領域に集合し、糸球クラスターを形成していること、および(3)糸球クラスターや糸球サブクラスターの空間的配置は一定で個体間で保存されていることが判明した。さらに、実験結果の詳細な解析から嗅球の外側半球地図内で「匂い分子受容体の受容サイト構造」が体系的に表現されていることが明らかになった。現在この「受容サイト構造地図」の空間配置の詳細を決定している。 (2)内因性信号の光学的測定法による嗅皮質からの匂い応答の記録嗅皮質は脳底部にあるため、これまで光学的測定法を用いることは困難であった。新しく開発した手術法を用い、ラットの嗅皮質の一部である前梨状葉から「匂い応答」を記録することに成功した。たとえばバナナのニオイで鼻腔を刺激すると、前梨状葉背側部の特定の領域に信号が観測され、この信号はさらに背側の島皮質(味皮質)へと広がった。この光学的測定法を用い、これまで全く未知であった前梨状葉における「匂い地図」を探索中である。
