哺乳類卵胞の選択的死滅制御機構とその人為支配による潜在的卵巣卵の利用

研究概要

サケのように一生に一度だけ全卵子を排卵する動物と異なり、哺乳類は性周期毎に一定数を成熟・排卵することを繰り返す。哺乳類の片側卵巣には、胎児期に減数分裂前期後半のディプロテン期で休止した卵母細胞が5〜50万個含まれている。出産後性成熟期に達すると、性周期毎に卵母細胞はそれを取り囲む卵胞の発達とともに発育する。この時卵巣内では最終的に排卵にいたる卵子の100倍以上の卵胞が発達する。この卵胞発達とそれに引き続いて起こる卵母細胞の成熟の過程で、99%以上の卵母細胞は選択的に死滅し、ごく一部の選抜された卵子のみが排卵にいたる。この卵胞の選択的死滅は、より優秀で強靭な子孫を残す戦略として重要であるが、この選択的死滅を制御している分子機構は未だ解明されていない。本研究は、未解明な哺乳類卵巣において発育・成熟中の卵母細胞を含む卵胞の選択的死滅を制御している分子機構を解明し、この知見をもとに細胞死を制御している遺伝子の発現をin vivo制御することで卵胞の発達を人為支配し、卵巣内に潜在する卵母細胞の有効利用を実現することが目的である。今年度は、新規な細胞死受容体を認識するユニークなモノクロナル抗体を作製し、それを用いて、新たに囮(デコイ)受容体を発見できた。また、卵巣で発現していることが知られている細胞死受容体であるFasの細胞内シグナル伝達系を担うカスパーゼをノックアウトしたマウスを用いて、卵巣におけるアポトーシスシグナル伝達系の分子制御機構を解明した。一方でウシ、ブタなどの大型哺乳類の未成熟な卵胞を免疫不全マウスの腎に移植(異種移植)し、発育・成熟する技術システムを確立した。
