研究概要	学術創成研究費「食物質による免疫作動機構の解明と応用技術の開発」で得られた平成13年度の研究実績は以下のとおりである。 1)マウス小腸粘膜の腸間膜反対側に100個から200個のB細胞集積を見出し、これらがマウスにおける孤立リンパ小節(isolated lymphoid follicles : ILF)であることを初めて報告した(J.lmmunol.168:57,2002)。この論文はNature ReviewImmunology(2 : 70,2002)にHighlightとして取り上げられ、注目すべき新発見として紹介された。 2)食物質/腸内常在細菌(フローラ)/腸管上皮細胞(IEC)/免疫担当細胞には機能的連結(相互作用)がみられ、フローラによる全身免疫システムの賦活は明らかであるが、その実体は未だ断片的である。本年度の研究によってフローラとB細胞による相互作用がIECの恒常性維持に重要であることを明らかにした(J.lmmunol.2002,inpress)。 3)腸管粘膜に分布する樹状細胞(dendritic cells ; DC)は食物質や腸内微生物に対する免疫応答の始動に重要である。インターロイキン15(IL-15)がDCの活性化によるIL-12産生に必須であることを確認するとともに自然免疫防御の始動(例:マクロファージによる病原細菌の殺菌)に極めて重要であることを明らかにした(Nature lmmunology 2:1138,2001)。 4)ストレスによって誘導されるHeme oxygenase(HO)はcatalaseやglutathioneの産生を介して酸化ストレスによる細胞傷害を防御することを明らかにする(J.Biol.Chem.2002,in press)とともに、HOによって産生されるCOがもっとも巧妙に利用され生体防御に関与すると考えられる下部消化管機能の研究を推進するために、ガス分子を用いた生体防御機構解明に必須の遺伝子改変動物リソースの作成を行った。
