研究概要

1、イネ耐病性シグナリングに関与するタンパク質を同定するため、活性型低分子Gタンパク質OsRac1を発現するイネ培養細胞からタンパク質を抽出し、2次元電気泳動で解析した結果、全部で173個発現の異なるタンパク質スポットを検出した。それらをさらにLC/MS/MSを用いて分析し、4個のタンパク質を同定した。
2、超乾燥・高温耐性の野生スイカの分子機構を解明するため、その遺伝子発現を解析し、メタロチオネイン(MT)と、新規2ヘム型シトクロムb(シトクロムb561)のmRNA蓄積量がストレスに伴い増大することが見出された。
3、アブラナ科植物の自家不和合性における自他識別が、花粉表層のSP11リガンドと柱頭細胞膜上のSRK/SLG受容体キナーゼ複合体との相互作用を介して行われていることを蛋白質レベルで明らかにした。
4、コムギChinese Springと1B染色体短腕の一部を欠失した系統から抽出した種子蛋白質を2-DEで分析した。次に、トリプシンで消化し、得られたペプチドをアフィニティークロマトグラフィーで精製した後、LC/MS/MSを用いて分析した。
5、植物を糖飢餓状態に置くと生命維持のためエネルギー生産に関わる機構が活性化する。即ちタンパク質や脂質の分解系や転流に関わる因子が誘導される。このようなタンパク質因子を同定するために35Sを取り込ませたシロイヌナズナから抽出したタンパク質を2D-PAGEで展開することによりタンパク質のプロファイリングをおこなった。
6、タバコ植物葉から抽出したタンパク質を二次元電気泳動で分離、抗ニトロチロシン抗体を用いてウェスタンブロットを行った。CBB染色で検出されるスポット4個のタンパク質について部分アミノ酸配列を決定した結果、酸性PRタンパク質のPR-1とPR-5(thaumatin-like proteins)であっ
