シロイヌナズナ完全長cDNAマイクロアレイを用いた遺伝子発現解析

研究概要	本研究では、モデル高等植物のシロイヌナズナを用い、シロイヌナズナ完全長cDNAマイクロアレイを用いた解析により、種々のストレス、ホルモンおよび光に応答する遺伝子等を単離、同定する。同定された遺伝子に関してプロモーター解析を行う。ビオチン化cap trapper法によりシロイヌナズナの完全長cDNAライブラリーをこれまでに19種類作製し、これまでに約155,144個のcDNAクローンを単離した。それらをクラスタリングしたところ、14,668個の独立したcDNAグループに分類された。これまでに単離した約7000個の完全長cDNAクローンを用いて完全長cDNAマイクロアレイを作製し、乾燥、低温、塩などのストレスおよび植物ホルモンABAに応答する遺伝子の同定を試みた。その結果、乾燥、低温、塩およびABA誘導性遺伝子はそれぞれ、299個、54個、213個、245個同定された。これら同定された誘導性遺伝子の中には転写因子と相同性を持つものが40個存在しており、乾燥、低温、塩、ABAのシグナル伝達経路には多くの遺伝子発現制御機構が存在していることが明らかとなった。同定された誘導性の転写因子の中には、NACファミリーやWRKYファミリーなどの植物特異的な転写因子も存在していた。また、ABA誘導性遺伝子の多くは乾燥および塩ストレス処理により誘導され、ABAと乾燥ストレスの両方により誘導される遺伝子は、ABAと低温ストレスの両方により誘導される遺伝子よりもはるかに多く存在していた。これらの結果は、これまでに考えられていたモデルと一致した。さらに、同定されたABA誘導性遺伝子のプロモーターについて解析したところ、ABA誘導性の遺伝子発現に関与する既知シス配列をプロモーター領域に持たない遺伝子が10個存在しており、ABA誘導性の遺伝子発現に関与する新規なシス配列の存在が示唆された。
