(DNAプール)PLACE-SSCP法による自己免疫疾患の病因遺伝子の網羅的探索

研究概要	免疫・アレルギー性疾患の候補遺伝子として、HLA領域をはじめとして、補体ならびにその受容体、T細胞受容体、免疫グロブリン受容体、各種サイトカインならびにその受容体、アポトーシス関連分子、オンコジーン、各種受容体の細胞内シグナル伝達分子など多岐にわたるが、HLA以外は充分な症例数について系統的検討がなされているとは言えず、特に日本人での大規摸な検討は皆無である。(DNAプール)PLACE-SSCP法は、数百人単位で迅速かつ正確にSNPを検出できるシステムであり、その特徴を利用して上記候補遺伝子の多型(SNP)ならびに変異の検索、膠原病・アレルギー疾患での頻度の解析、疾患との関連を解析する。我々は今までに、Dnase 1、caspase-8、APRIL、Fas、HS1などの分子の異常あるいはポリモルフィズムとSLEの関連について報告してきた。その結果、アポトーシスに関連した分子の機能異常がSLEの病態に大きく関与していることが示唆された。今年度は、疾患モデルマウスで異常が見い出された分子や、アポトーシス関連分子、TNF/TNF受容体ファミリー分子を中心として40個の遺伝子を選択し、全エクソン領域についてSNPの有無、その頻度をSLE患者、健常人で検討した。さらに機能解析にも着手している。我々の開発した画期的な遺伝子解析システムによりさらに効率よく網羅的な疾患関連遺伝子を同定できると考える。
