植物の光合成遺伝子システムに特有なコアプロモーター構成の起源と役割

研究概要

植物の核遺伝子の多くはコアプロモーターの成分としてTATAボックスをもっているが、光合成遺伝子群にだけはTATAボックスがあまりみられない。光合成核遺伝子の大部分はもともとは原核型の葉緑体ゲノムにコードされていたものが、進化の過程で核ゲノムに転移し、そこで新たに真核型のプロモーターを獲得したと考えられている。本研究では、光合成遺伝子群が特有なコアプロモーター構成をもつ理由を、その起源と役割の二つの側面から検討する。本年度は、光合成遺伝子プロモーターについて、(1)構造的特徴の検討、(2)キメラプロモーターを用いた機能解析、(3)核ゲノム中でのプロモーター獲得過程を再現する実験系の確立、の3点を中心に研究を進めた。主な成果は以下のとおりである。
(1)光合成膜タンパク質遺伝子のプロモーターをシロイヌナズナやイネを材料に詳細に解析した結果、光合成装置は、葉緑体ゲノムにコードされている反応中心サブユニットの周囲をTATA-less型核遺伝子の産物が取り囲み、さらにその周辺部にTATAボックス型核遺伝子の産物が位置している、という階層的な構造をもつことが明らかになった。
(2)光合成遺伝子をはじめとする植物遺伝子群からコアプロモーター領域と調節プロモーター領域を取り出し、それらを組み合わせたキメラプロモーターの機能をタバコやイネで解析した。その結果、植物の核遺伝子のコアプロモーターと調節プロモーターの間には、少なくとも3種類の適合性群があり、その群を超えた組み合わせでは適切な転写調節が生じないことが明らかとなった。この知見は、コアプロモーターの類型は遺伝子の発現制御機構と深い関わりのあること示している。
(3)シロイヌナズナの遺伝子トラッピング系統約350について外来遺伝子の挿入部位や発現の有無を解析し、核ゲノム中で偶発的に生じる新規プロモーターの構造的特徴を明らかにした。
