SHIV-サルの系を用いたエイズウイルスの粘膜感染機序と粘膜防御機構の解析

研究概要

HIVの粘膜感染機序と粘膜防御機構の解明の為に、SHIVの経粘膜感染ザルを用いて、特に感染初期のウイルス増殖機序と免疫誘導機構の解析を個体レベルで実験的に明らかにすることを目的として、今年度は以下について重点的に検討した。
1.全身臓器におけるウイルス動態
ウイルス接種3日後に、プロウイルスDNAはリンバ節、胸腺等からも検出され、速やかに全身へ拡散することが明らかになった。13日後に、リンパ系組織中にウイルスが検出されたが、27日後にはプロウイルス量が高いにも関わらず感染性ウイルス量は激減し潜伏感染となっていた。一方、腸管においては、高いプロウイルスDNAにも関わらず、感染性ウイルスはほとんど検出されず、早期から潜伏状態である事が明らかになった。
2.腸管粘膜における免疫応答
腸管のCD4+細胞数は13日まで変化は認められなかったが、27日には1%に激減した。またCD4陽性細胞の減少に先立ってCD4+CD8+細胞が減少することが明らかになった。腸管組織内のγδT細胞は、感染後6日に一過性の増加が見られた。さらに、末梢血やリンパ節におけるNK活性は低かったが、腸管は非常に高いNK活性を示した。
3.胸腺におけるT細胞分化障害
胸腺内で13日に組織の荒廃とCD4+細胞の減少が特に著しかったが、それに先立ってCD4+CD8+細胞(DP)が減少していた。さらに胸腺内T前駆細胞(CD3-CD4-CD8-)のex vivoにおける胸腺分化システム(FTOC)を行ったところ、6日以降のDPの割合は非感染サルから採取したものに比べ減少していた、このことは胸腺におけるT細胞分化障害がex vivoの実験で明らかになり、CD4+細胞枯渇の原因の一つであることを実証した。
