細菌性腸管感染症の初期過程(細胞への付着と侵入)に関わる因子と細胞応答反応

研究概要	本研究では本田が生物学的な解析を進めると共に研究を総括し、遺伝学的解析に実績のある柳原が、2名のポスドクと13名の大学院生を指導しながら、研究を進めた。(1)腸管出血性大腸菌に関する研究:EHECあるいはEPEC由来の精製Tir(付着のため菌自身が用意するIntiminレセプター)を用いたアフィニティカラム、抗Tir抗体を用いた免疫沈降法などでTirと結合する生体分子としてCortactinとTalinをこれまでに見出してきたが、さらにこれら2分子はそれぞれ直接Tirに結合することをLigand-overlay法などで明らかにするとともにこれら2成分はEHEC感染部位のアクチン凝集に関与していることが示唆できた。さらに、EHECの3型分泌装置を通じて標的細胞に注入されると考えられているエフェクター分子の1つであるEspBがαCateninと結合する可能性を見出した。(2)腸炎ビブリオに関する研究:腸炎ビブリオの腸管内定着に関与する因子の同定を目指しこれまでの研究で、菌側の因子(蛋白)としてcHA、遺伝子としてvagを見出したが、両者には現在のところ、関連性が見出せない。したがって本菌の付着には、複数の物質が関与していると考え、さらに解析を加えている。(3)Providenciaに関する研究:2001年に我々が見出した新型食中毒菌providencia alcalifaciensが細胞侵入性を持つことが分かったが、細胞侵入性を示さない菌株も存在したので、プラスミドの有無と侵入性の関係を解析したところ、約150kbp大のプラスミドの有無と相関する事が分かった。現在、このプラスミドを解析中。
