ガレクチンファミリーを介した自然免疫系の活性化と感染応答の分子基盤

研究概要	ガレクチンファミリーは、βガラクトシドを特異的に認識する動物レクチンで、脊椎、無脊椎動物に広く分布している。しかし、ガレクチン分子種の詳しい組織分布や機能についてはほとんど分かっていない。本研究では、感染応答におけるガレクチンファミリーの役割、中でもタンデムリピート型ガレクチン(galectin-4、-8、9など)を中心に、免疫系細胞の活性化と糖鎖認識を介した情報伝達の分子基盤の解析を行う。我々は昨年度、galectin-8の二つの糖認識ドメイン(CRD)のうち、N-末端側CRDはヒト好中球のproMMP-9と、C-末端側はproMMP-9とintegrinαMと結合することを明らにした。本年度、galectin-8が血液凝固因子トロンビンによって、2つのCRDに切断されることやその切断部位を明らかにした。また、MMP-3によるproMMP-9の活性化が、galectin-8存在下では顕著に促進されることを見出した(論文投稿中)。以上の結果は、細菌や異物の感染に伴って、生体内のgalectin-8が好中球を活性化し得ることや、感染に伴って起こる炎症や好中球の組織への浸潤の調節にgalectin-8が関与している可能性を示唆している。一方、炎症性サイトカインであるインターフェロンγ刺激で、galectin-9が血管内皮細胞、繊維芽細胞などで特異的に誘導されることや、培養細胞(Jurkat T細胞)に対して、galectin-9がアポトーシスを、galectin-8は細胞接着を誘導することも見出している。また、galectin-8、-9の認識する糖鎖構造の特異性をフロンタルアフィニティークロマトグラフ法により明らかにした。このように、感染や炎症部位の免疫細胞の接着や浸潤において、糖認識を基盤にして、タンデムリピート型ガレクチンを介する全く新しい経路が機能している可能性がある。
