異物排出システム群の連携作動解析による多剤耐性緑膿菌感染症の新規化学療法の開発

研究概要	緑膿菌による高齢者や免疫不全患者での重篤な難治感染症における抗菌薬耐性の軽減を目指し、異物排出に働くマルチコンポーネント型排出システムの機能および発現制御機構を解析し、下記の結果を得た。 1.基質認識機構:基質認識の本体であることを明らかにしたRNDコンポーネント分子内の基質認識アミノ酸を解析したところ、E.coliのRNDであるAcrBのX-線結晶のポケット構造に相当する領域に基質認識に寄与するPheクラスター構造が存在することが分かった。 2.抗菌薬耐性および病原性への関与:病原性に関与する代謝産物の細胞外輸送の可能性を調べた。MDCK細胞モノレイヤ透過実験系から、MexAB-OprM排出システムの欠損は細胞侵入性を失わせることが分かった。これは白血球減少マウスでの病原性の喪失と一致した。 3.発現制御機構:緑膿菌シグマ因子σDとσSタンパク質を大量精製し、試験管内での転写系、DNA-タンパク質結合反応、in vivoプロモーター活性を測定することにより、現在ゲノム上で確認されている12種類の排出系遺伝子プロモーターのシグマ因子特異性の決定を行い、転写制御機構と転写制御機構全体像の解明を行っている。排出システム阻害薬のデザインを目指した基質認識の分子機構の解明と緑膿菌細胞での排出システムの発現と機能の解明の二面から研究を進めてきた結果、1)阻害薬のデザインのための分子構造に関する有用な情報を提供することができた。2)排出システムが病原因子の排出にも機能していることから、排出システムの機能の阻害は抗菌薬耐性の軽減のみならず、緑膿菌の病原性の低減にも繋がることが分かった。このように、多種の排出システムを秘めた緑膿菌で、排出システムの機能の網羅的な解明は本菌感染症の新規化学療法を開くものと期待される。
