持続性に優れたワクチン開発のための記憶B細胞長期生存に関わる因子の同定

研究概要	持続性に優れたワクチン開発のための記憶B細胞長期生存に関わる因子の同定のために、記憶B細胞で高発現する膜蛋白・分泌蛋白をコードする遺伝子のスクリーニングを行った。すなわち、記憶B細胞から大量にcoding RNAを合成し、大規模なレトロウィルスcDNAライブラリーを作成した。これを用いて、シグナルシークエンストラップ法によるスクリーニングを行った結果、約2000の膜蛋白・分泌蛋白をコードするcDNAクローンの単離に成功した。得られたクローンを用いてcDNAマイクロアレイを作成し、記憶B細胞と未感作B細胞での発現量を比較したところ、3倍以上の変化が見られたクローンを59個検出した。これとは別にマウスの約8600の遺伝子を含むcDNAマイクロアレイに対して、記憶B細胞及び未感作B細胞より精製したRNAを増幅してプローブとしてハイブリダイゼーションを行うたところ、マイクロアレイ上記憶B細胞で2倍以上発現の変化する遺伝子として440個の遺伝子を同定した。この中には判明しているだけで78の膜蛋白・分泌蛋白が含まれており、そのうち特に、RT-PCR法によって記憶B細胞で高発現を確認した既知のケモカインレセプターをコードするclone Ag2、同じく既知のサイトカインをコードするclone Ag190など、6つの遺伝子について注目し、記憶B細胞における蛋白としての発現や機能について現在解析を行っている。
