研究概要

DNAのメチル化はエピジェネティックな遺伝子発現調節機構の一つであり、細胞のがん化、分化などに重要な役割を担っていると考えられている。その代表的標的配列cAMP responsive element (CRE)は多くの遺伝子のプロモーター上に存在し、cAMPによる遺伝子発現調節にて重要な役割を担っている。本研究では、メチル化されたCREに結合する因子としてMBD2に着目し、メチル化CREにおける転写制御機構の解析を試みた(MBD2はスプライシングによりMBD2aとMBD2bの二つのアイソフォームからなる転写抑制因子である)。MBD2a, MBD2bはともにメチル化されたCREの転写を抑制した。一方で、予想に反して、MBD2aのみがメチル化していないCREからの遺伝子発現を活性化した。結合実験により、MBD2aはCREB転写装置の構成因子であるRNAヘリケースA(RHA)と選択的に結合し、さらに、MBD2aとRHAは協調的にCREB依存的な転写を活性化した。また、RHAと結合しているMBD2aは、転写コリプッレサーであるHistone deacetylase 1とは複合体を形成していなかった。以上の結果から、MBD2aは転写抑制複合体と相互作用して転写を抑制するだけでなく、転写活性化複合体としても作用しうる可能性が示された。
