発癌過程におけるコネキシン機能異常のメカニズム;細胞接着因子との相互依存性

研究概要

癌細胞のほとんどでギャップ結合コミュニケーションが低下・欠乏しているが、その中でもコネキシンの細胞内局在異常が多いことを考え、コネキシンの局在性を制御するタンパクの存在を仮定した。そこで、Cx26と相互作用するタンパク質(AP26)をHeLa細胞のcDNAライブラリーから酵母two-hybrid法で同定した。AP26遺伝子はほぼ全ての組織で発現していたが、Cx26が細胞膜に局在する細胞でのみAP26も細胞膜に存在した。
AP26の発現をsiRNAで阻害した細胞では、Cx26の局在が細胞膜から細胞質内へ移動し・細胞間コミュニケーションも阻害された。さらに、AP26が結合するCx26の細胞質内ループのみを過剰発現させた細胞でも、Cx26は細胞質内へ移行し、コミュニケーションが減少した。これらの結果は、AP26がCx26の細胞質内ループドメインに結合して裏打ちタンパク的役割を果たしていることを示唆している。Ap26がアンドロジェン・リセプターの共転写因子として作用する。という報告から考えて、AP26はWNT情報伝達系におけるβ-カテニン的役割を果たしている可能性が示唆される。Cx43にも同様なタンパク(AP43)が存在すると想定して、酵母two-hybrid法で4個の候補遺伝子をクローニングした。AP26とAP43の機能を平行して解析することによって、コネキシンの局在性および情報伝達系が解明される可能性がある。
