ヒトパピローマウイルスによる発がん機構の解析

研究概要	HPV陽性の子宮頸がん細胞株にレトロウイルスベクターを用い、ウイルス転写制御因子E2を高率に導入しE6とE7の発現をほぼ完全に抑えることができる系を確立した。E2導入により上記HPV陽性の子宮頸がん細胞株は増殖を完全に停止するのに対し、HPV陰性の子宮頸がん細胞株C33Aでは増殖に全く影響を与えない。このとき、HPV陽性の子宮頸がん細胞株ではp53の蓄積、p21WAF1の誘導、低リン酸化型RBの増加が見られ、細胞周期はG1で停止した。次にHPV18のE6とE7を発現しているHeLa細胞にあらかじめHPV16のE6とE7を単独あるいは組み合わせてレトロウイルスベクターを用い導入しておくことでHeLa細胞の増殖のE6とE7に対する依存性を区別して解析することができた。その結果、E2導入による細胞増殖停止は主にE7の発現消失に伴う低リン酸化型RBの増加によるものでRBのリン酸化阻害は既に高発現しているp16^<INK4a>によるものであることが示唆された。現在HeLa細胞の造腫瘍原性におけるE6の役割を解析中である。これまでにE6を発現しないHeLa細胞をヌードマウスへ移植すると造腫瘍能が消失するとの結果を得ている。一方、子宮頸部正常上皮細胞ががん化に至る過程を解析するため、同細胞をE6E7の導入により不死化することに成功している。さらに、不死化からがん化に至る過程でのE6とE7の機能を解析するためには、E6E7を導入せずに不死化した細胞にE6、E7を導入してそれによる変化を調べることが必要である。これまでにhTERTとbmilを導入することで同細胞を不死化することに成功している。現在この細胞にE6,E7の他ras^*などのがん遺伝子を導入しがん化に必要な条件を検討中である。
