Drs遺伝子機能と足場非依存性増殖の分子機構

研究概要	我々が分離した癌抑制遺伝子Drsはヒト癌細胞株においてサイクリンAの発現抑制を介して足場非依存性増殖を抑制する。また、我々は大腸癌、肺腺癌、前立腺癌などのヒト悪性癌組織でdrs mRNAの発現抑制が高頻度で起こっていることを明らかにしてきた。本年度、我々は主にプロテオミクスと発生工学の手法を用いてDrs遺伝子の機能と癌化抑制機構を解析し以下の成果を得た。 1.ATLのリンパ腫組織でHTLV Taxの発現に依存しないdrs mRNAの発現抑制が高頻度で起こっていることを見い出した。またATL由来細胞株にDrs遺伝子をレトロウイルスベクターで導入すると増殖抑制を示すことを明らかにした。 2.プロテオミクスの手法を用いて同定した複数のDrs結合蛋白の中でGRP78/BiPは小胞体においてストレスによるアポトーシス誘導に関わっている。Drs遺伝子とアポトーシスとの関連を検討するために種々のヒト癌細胞株にDrs遺伝子を高発現させたところ、Drs遺伝子がCaspase-3とCaspase-9の活性化を伴うアポトーシスを誘導する活性を持つことを見い出した。またDrsはアポトーシス誘導蛋白ASY/Nogo-B/RTN-Xとも結合することも明らかにした。 3.ジーンターゲティング法によってDrs遺伝子ノックアウトマウスを作製することに成功した。現在までに雄(-/Y)と雌(-/-)の両方のDrs KOマウスが胎生致死にならずに生まれてきており、これらのマウスは生殖能力も保持しており交配可能である。これらのDrs KOマウスとそれに由来する初代培養細胞を用いてDrsの個体レベルでの機能と癌化形質の発現に対する影響、また細胞レベルでの増殖、不死化、癌化、アポトーシス、接着、分化などにおけるDrs遺伝子の役割を検討中である。
