新規がん転移関連遺伝子(タンパク質リン酸化酵素BRPK、糖転移酵素3H8)の解析

研究概要	がん転移の分子機構を理解し、その制御方法を開発するため、胎盤形成と転移能の異なるがん細胞株における発現変化を指標に単離した新規遣伝子BRPKと3H8を解析した。 1.タンパク質リン酸化酵素BRPKの解析組み換えBRPKタンパク質は、試験管内で自己リン酸化能を示すと同時にヒストンH1をよい基質とした、細胞内では、細胞質と核の両方に存在する。結合タンパク質をスクリーニングした結果、PCNAとBRPKのkinase domain領域が結合することが示唆された。細胞抽出液の免疫沈降でもBRPKはPCNAと共沈し、細胞をaphydicholinで処理すると共沈するBRPK量が増加した。免疫染色でも、aphydicholin処理時に核内におけるBRPKはPCNAと共局在する程度が増加した。更に、PCNAは試験管内でBRPKのリン酸化活性を抑制した。以上から、BRPKは細胞周期依存的にPCNAと相互作用をする可能性が示唆された。 2.糖転移酵素3H8の解析アミノ酸配列ホモロジーの系統樹解析から、3H8はβ1, 3-N-acetylglucosaminyltransferaseの一種であると推定された。マウス各臓器では、胎盤と大腸で発現が高く、肺、小腸、腎臓でも発現が見られた。アンチセンスオリゴで発現を抑制すると、in vitroで細胞の遊走能や浸潤能が亢進した。従って、3H8は細胞遊走・浸潤に抑制的に作用していると考えられる。以上の結果は、今後検討すべき課題は多いにせよ、2つの新規遺伝子がヒトがんの増殖異常、浸潤転移の機構を理解しその対策を講じる上で有用であることを示す。
