サイトカインシグナルを分子基盤とした癌細胞の増殖制御機構の解析

研究概要	生体内各器官の形成と維持においては、それらを構成する種々の細胞群の増殖と分化がサイトカインなど細胞外来性のシグナルやエピジェネティックな修飾を基盤とする細胞内在性のプログラムにより精妙に制御されることが必要であり、その破綻が発癌に至ると考えられる。本研究は、細胞増殖を正に制御するサイトカインシグナルと負に制御するシグナルの相互作用に焦点をあてた癌細胞の増殖制御機構の解明を目指した。特に、研究代表者らが取り組んできたサイトカイン受容体コンポーネントgp130を刺激するoncostatin M(OSM)と相乗作用して細胞増殖を誘導することが知られているstem cell factor(SCF)やbasic fabroblast growth factor(bFGF)などのサイトカインシグナル経路に対してシグナル抑制性分子Spred-2やLnkあるいは白血病関連分子fused in myeloproliferative disorder(FIM)などがどのように相互作用して細胞の増殖応答を決定するか解明を図った。OSM、SCFおよびbFGFの3つのサイトカインで血球が出現する胎生期マウス造血組織であるaorta-gonad-mesonephros(AGM)領域のin vitro培養系を用いてこの点に取り組んだ結果、(1)AGM造血期に発現を確認したRas/MAPK経路を負に制御する分子であるSpred-2がAGM領域からの造血前駆細胞集団の拡大を負に制御していることがわかった。(2)SCFの受容体c-Kitに結合するアダプター分子として知られているLnkがAGM領域における造血を制御することを見いだした。また、(3)白血病においてキメラ分子を作るFIMがAGM領域における造血前駆細胞集団の拡大を抑制することもわかった。サイトカインシグナルの破綻による細胞の異常な増殖が発癌に至る機構の一つであることを考慮すると、本研究の知見は発癌の仕組みの解明ならびにその抑制法の開発に関して興味深い示唆を与えるものと考察される。
