研究概要	NF-κBサブユニット遺伝子の増幅および遺伝子変異がリンパ腫や胃癌などで報告され、またHTLV-1やEBVなどのウイルス発がんにNF-κBが関与するなど、両者の因果関係が多くの事実から示唆される。また、がん転移に関わる細胞接着因子(E-selectin,ICAM-1,など)がNF-κBによって転写制御を受ける。本研究では、ヒトゲノム計画の成果のひとつとして新たに見つかったBlyS/BAFF/TALL-1および同じシグナル伝達系を使用してNF-κBを活性化するLTシグナルの詳細を調べた。今年度の本研究で以下の事実を明らかにした。(1)LTbRシグナリングでのNF-kB活性化はIkBリン酸化およびp65の核移行に依存しない。(2)様々のNF-kB活性化因子とそれらのdominant negative変異体を導入し、Luciferase assayでその作用を解析した結果、TNFシグナリングは主にIKKb、すなわちIkBリシ酸化cascadeを使うが、LTbRシグナリングは主にNIK、IKKa cascadeを使っていることがわかった。(3)NIK-IKKaの作用機構をさらに確認するため、p65wild-typeと様々の変異体のGal4融合蛋白の発現プラスミドを作成してNIKにより誘導される転写活性を比較した結果、p65のTA1領域とりわけTA1領域の536番セリンが必須であることがわかった。(4)実際にLTbRシグナリングにおけるp65の536番セリンのリン酸化を証明した。これらの成果は、発がんおよびがん進展におけるNF-κBの役割とその分子機構についての理解を一歩すすめ、新たな治療法開発に重要な情報を提供した。
