研究概要	4F2hc(4F2抗原重鎖)は、リンパ球の活性化抗原及び腫瘍抗原として同定され、細胞の形質転換及び増殖制御との関わりが想定されている1回膜貫通型のII型膜糖タンパク質である。本研究は、4F2hcを介するシグナリング機構を明らかにする目的で、4F2hcの細胞内ドメインに連結するタンパク質を酵母two-hybrid法を用いて探索し、細胞増殖及び形質転換に関わるシグナル経路との関連を検討した。その結果、FKBP12.6及びUbc9が酵母two-hybrid法により陽性クローンとして得られ、4F2hcの細胞内ドメインに連結するタンパク質との候補として挙げられた。さらにT24細胞及びNIH3T3細胞において、4F2hcの強制発現により、p70 S6kinaseとAktのリン酸化が充進し、mTOR経路の活性化とともに、PI3 kinaseの活性化が生じることが示唆された。これに対して、4F2hcとヘテロ二量体を形成することによって機能するアミノ酸トランスポーターLAT1(L-type amino acid transporter 1)を介するアミノ酸取り込みの抑制によっては、mTOR経路のみが影響を受け、PI3 kinase経路はこれに関わらないことが明らかになった。従って、4F2hcは、アミノ酸トランスポーターの補助因子としてアミノ酸輸送を上昇させる他、アミノ酸トランスポーターを介さずにそれ自体が細胞内シグナル機構と連結することが示唆された。本研究の成果は、4F2hcを介するシグナリング機構の研究の端緒を開くとともに、mTOR経路の上流因子に関する新たな示唆を与えるものである。
