研究概要

Axamは私共が見出したAxin結合蛋白質588アミノ酸からなる蛋白質である。Axamは中央(72-400)にAxin結合部位、C末端側は脱SUMO化酵素SENP1と相同性が高い部位を持つことが明らかとなった。SUMOはユビキチン類似の分子で、蛋白質の細胞内局在や安定性の制御、転写活性の調節などに関与することが知られている。実際に、AxamはSUMO化されたp53に対して脱SUMO化活性を示し、COS細胞にAxamをSUMO-1と共発現させると他の蛋白質と共有結合しているSUMO-1が限定分解されてモノマーとなることが明らかとなった。したがって、Axamは細胞内でも脱SUMO化酵素として作用すると考えられた。Axamはβ-カテニン分解を促進し、Wntシグナル伝達経路依存性の体軸形成を抑制するが、脱SUMO化酵素活性を欠失させたAxamC547SではAxam全長で認められるβ-カテニン分解作用とWntシグナル依存性の二次体軸形成の抑制作用が低下した。したがって、AxamによるWntシグナル伝達の抑制にはAxamの脱SUMO化活性が重要であることが示された。
Duplinは私共が見出したβ-カテニンと結合し、初期胚の体軸形成やWntシグナル伝達を抑制する核内蛋白質である。変異体解析の結果、Duplinのアミノ酸残基(500-584)が本蛋白質の核内局在に必要かつ充分であり、Wntシグナル依存性のTCF転写活性化の阻害にも必須であることが明らかとなった。また、DuplinのC末端側のβ-カテニンと結合部位を欠失させた変異体Duplin-(1-668)はβ-カテニンと結合せず、Wnt3a依存性のTCF転写活性化の抑制能も失われていた。ところが、アフリカツメガエル初期胚の背側にこの変異体Duplin-(1-668)を導入したところ、Duplin全長と同様の体軸形成抑制作用を示した。したがって、Duplinはβ-カテニン以外の下流の遺伝子発現のシグナル伝達経路にも作用することにより体軸形成を抑制する作用を持つことが示唆された。
