翻訳マシーンの分子擬態とプリオン特性の研究

研究概要

分子擬態に関する研究
1.ペプチド解離後のリボソーム複合体は再生因子(RRF)と翻訳伸長因子EF-Gの働きにより解体・再生される。RRFタンパク質はtRNAと構造が酷似するため、RRFはtRNAの同じリボソーム部位にエントリーして、tRNAと同様にEF-Gによってリボソーム上で押されて移動し、複合体から解離するというモデルが梶らによって提唱されていた。しかし、EF-GのtRNA転移機能不全変異はRRFの機能に何ら障害を起こさず、モデルの誤りを証明した。
2.これまで構造が明らかでなかった真核生物の2型解離因子eRF3の結晶構造を分裂酵母eRF3を持ちいて世界で初めて明らかにした。同時にGTP結合、GDP結合、非結合の3つの構造を明らかにした結果、通常のGタンパク質と異なり、GTP/GDP間でコンフォメーションの変化は観察されなかった。1型解離因子eRF1の結合部位を変異体の活性測定により明らかにした。
3. 23SrRNA変異体リボソームを利用した試験管内ペプチド解離分析によって、23S rRNA上のA2602塩基が加水分解のRNA触媒部位として働く可能性が明らかになった。
酵母プリオンに関する研究
1.酵母eRF3はSUP35遺伝子によりコードされ、そのN末端ドメインのプリオン性ペプチド配列により動物プリオンと共通する特徴を示す。Sup35プリオンの機能不全は終止信号無視である。N末端のプリオンドメインはGln/Asn領域とペプチド反復領域に分離されるが、ペプチド反復配列からGln/Asnを完全に除去した変異体を作成し解析した結果、既存のSup35プリオンとは異なる(シャペロンHsp104非依存性)新種プリオンとなることが分かった。
2.リピート構造を人為的に改変することによってプリオン化の程度が異なる各種プリオン「種」が生成され安定に継代することを発見した。これらは遺伝学的にはプリオンの「strain」として定義できる。
3.異種Sup35プリオンは種間障壁(species barrier)により伝搬が阻止される。この種特異性がN末端40アミノ酸領域に規定されることを発見した。
