細胞壁構築遺伝子群の総括的発現解析による軸性形成の制御機構の解析

研究概要	植物の軸性の基盤となる安定な細胞極性の成立と維持において中心的な役割を担う細胞壁に焦点を当て、その構築に関わる762種の遺伝子について、各組織内での発現パターンを体系的に解析し、その発現制御に係わる制御機構の解明を最終目的として研究を進め以下の成果を得た。 1)シロイヌナズナの細胞壁構築に関連した768遺伝子(28種の遺伝子ファミリー)を選抜し、個々の遺伝子を特異的に識別可能なオリゴDNAプローブを設計し、それを打ち付けたDNAマイクロアレーの解析法を確立した。 2)シロイヌナズナ花茎の表皮細胞の長さの分布と、成長に伴う分布の変化を解析した。この系を用いて、花茎の細胞伸長中および、細胞伸長後に発現の高まる細胞壁構築遺伝子群をそれぞれ特定した。 3)ブラシノステロイド合成に係わるDIM遺伝子を欠損したdiminute変異体を熱処理によりコンディショナルに相補できる形質転換体を用いて、ロゼッタ葉展開過程に増減する細胞壁関連遺伝子群をDNAマイクロアレー法を用いて特定した。 4)根特異的発現細胞壁関連遺伝子群をDNAマイクロアレー法により特定した。 5)茎の伸長、葉の展開、根の伸長に、それぞれ係わる遺伝子群は、それぞれの器官に特異的であることが明らかとなった。 6)根で特異的に発現するAtXTH17,AtXTH18,AtXTH19,AtXTH20それぞれの遺伝子について、1k Pro::GUS,0.3kPro::GUS,0.1Pro::GUSを導入した形質転換体を用いて4つの遺伝子のプロモーター解析を行い根特異的発現に係わるシス領域を特定した。
