研究概要	1.DJ-1の抗酸化ストレス機能 DJ-1は過酸化水素などの活性酸素により誘導される、還元型から自己酸化されることで活性酸素を除去することがin vitro, in vivoで明らかとなった。この機能にはDJ-1のダイマー形成が必須である。6-OHDA,MPP+などの神経毒は細胞に活性酸素を誘発し酸化ストレスを起こすことで神経細胞死を起こすが、DJ-1ノックダウン細胞ではこれらに対する感受性が増加し、逆にDJ-1過剰発現細胞では抵抗性を示した。DJ-1のSUMO-1化、ダイマー形成、DJ-1プロテアーゼ能の変異体、パーキンソン病に見られるL166P変異では自己酸化能、細胞死抵抗性が欠如していた。 2.ビスフェノールAによる活性酸素誘導とそれに対応するDJ-1の誘導ビスフェノールAはエストロゲン様活性により内分泌かく乱作用を有すると一般に考えられているが、エストロゲン活性は極めて弱く、ビスフェノールAの多様な作用機作は解明されていない。ビスフェノールA投与マウス脳、精子、培養脳、精子関連細胞では活性酸素が誘発され、それに伴ってDJ-1の酸化体と発現誘導が起こった。ビスフェノールA投与マウスはパーキンソン病様の神経細胞死が起こることが知られているが、1と合わせて酸化ストレスがビスフェノールAの1つの作用機作と考えられ、その除去にDJ-1が関与することが明らかとなった。 3.DJ-1のSUMO-1化とプロテアーゼ機能 DJ-1の130番目のリジンがSUMO-1化されることがDJ-1の全ての活性に必須である。L166P変異は過剰にSUMO-1されることで機能不全を起こしていた。方、DJ-1はC106を活性中心としたプロテアーゼであり、その基質としてパーキンソン病で封入対を形成するPaer受容体を同定した。
