新規興奮アミノ酸受容体遮断薬の全合成研究

研究概要	カイトセファリンは、興奮性アミノ酸受容体に対して高選択的な遮断活性を示すことから、神経機能研究のための手段物質や脳保護薬創製のためのリード物質の開発の見地から全合成研究が取り組まれている。その構造は、ピロリジン環の両端にセリン及びアラニンが炭素・炭素結合で連結し、特にそのセリン部位がピロリジン環と4級炭素結合型アミン構造を形成していることが特徴的である。そこで、鍵となる4級炭素を含む光学活性α-ヒドロキシグルタミン酸ユニットは不斉転写型ストレッカー合成で構築し、その脱水環化体の両端にアミノ酸を導入してカイトセファリンに導く合成経路を立案した.すでに、重要中間体であるα-ヒドロキシメチルグルタミン酸の合成経路が確立したことから、本年度は右側鎖の構築について検討した。分子モデルの考察からヒドロキシメチル部位の不飽和エステルへの変換と、引き続く段階的なアミノヒドロキシ基の導入が天然品の立体配置を満足する生成物を与えることが推定された。そこで、2段階でアルデヒド体に導き、安定イリドとの反応によりE-不飽和エステルに導いた。オスミウム酸化は立体選択的に進行し、C2',3'位がsyn配置のジオール体を与えた。C2'位へのS_N2型のアミノ基の導入に際して、アミドの保護基による立体遮蔽や水酸基の脱離などの問題に直面した。この問題は、アミド基が無保護の基質を用いて反応を行うことで解決できた。ジオールのチオカルボネート体はアジドと反応して、望ましい生成物を与えた。3'位の水酸基をTBS基で保護し、アジド基を還元したところスピロビスラクタムを与えた。アミノヒドロキシ基の相対立体配置はNOE実験から望ましい構造をもつことが確認できた。右側鎖の導入による全合成の完成にむけて、研究をすすめている。
