糖脂質糖鎖による膜ミクロドメイン異常

研究概要	糖脂質糖鎖の生体内役割を解明するために、現在解析中の2種のノックアウトマウスに加え、ラクトシルセラミド合成酵素であるβ 4GalT-6、GM3合成酵素、およびGb3/CD77合成酵素の遺伝子ノックアウト(KO)マウスの作成を進めた。 β 4 GalT6のKOマウスは生後1年近く経過しているが明らかな異常を認めない。mRNA発現はノザンブロットでもリアルタイムPCRでも完全に消失しているにも関わらずガングリオシドの構成に変化が見られず、現在他のβ 4 GalT familyのメンバーの発現を検討中である。糖脂質糖鎖が細胞膜ミクロドメイン(ラフト)において種々のシグナルの調節に関与する時のメカニズムを解明するために、糖鎖遺伝子で糖鎖リモデリングを行った細胞に関するシグナル変化とラフトの変化を検討した。PC12細胞にGM1合成酵素遺伝子を導入した時のNGFシグナル抑制について以前にも報告してきたが、そのメカニズムとして、TrkA、p75^<N7R>、Rasなどのラフトから非ラフトへの移行が認められた。一方、EGFに対するシグナルの検討では、GM1の高発現により増殖シグナルが亢進すると共に、ECF受容体のラフト/非ラフト局在がほとんど変化しないことを見い出した。さらに、GM1とこれらの受容体との会合を免疫沈降によって検討した結果、TrkAは強く結合するのに対して、EGFRは極めて微量しか結合していない事が示された。これらの結果は、GM1高発現の影響の程度は、受容体の元来の局在とGM1の結合性の強さに依存することが示された。一方、メラノーマなどの神経外胚葉性腫瘍によく発現するGD3、GD2などの、増殖、転移などの悪性形質における意義につき、糖鎖リモデリング細胞を用いて解析した。GD3発現は、メラノーマにおけるEGF、PDGF、bFGFなどの増殖シグナルについては大きな変化を与えず、血清刺激による増殖シグナルのみを大きく増強した。その機構として、FAK、p130Cas、MAPK、またはAktというシグナル経路の活性化を惹起することが示された。現在、ラフトにおいてGD3と直接会合する分子の検討を進めている。また実際のメラノーマ組織におけるこれらのシグナル系の活性化につき検討の予定である。
