神経系構築における硫酸化ヘキソサミン識別

研究概要	N-アセチルグルコサミン6-スルホトランスフェラーゼ(GlcNAc6ST-1)遺伝子のノックアウトマウスについて研究を進めた。ノックアウト胚の視床下部では、ケラタン硫駿が欠失し、GlcNAc6ST-1がこの部位でのクラタン硫酸の合成に関与することが明らかになった。また、このケラタン硫酸を持つプロテオグリカンを追跡して、受容体型チロシンホスファターゼζ(PTPζ)がその一つであることを見出した。現在、GlcNAc6ST-1ノックアウトマウスにおいての神経機能の変化を解析している。GlcNAc6ST-1ノックアウトマウスでのL-セレクチンリガンドの発現状況についても研究した。ノックアウトマウスでは、ノイラミニダーゼ消化後のAG223抗原の発現がリンパ組織で大きく低下し、L-セレクチンリガンドの合成に関与することが明らかとなった。一方、MECA-79抗体による染色では、アブルミナミルなリガンドの低下が著しかった。GlcNAc6ST-1ノックアウトマウスでの、リンパ球ホーミング能についても解析した。すると、末梢リンパ節、腸管膜リンパ節、パイエル板へのホーミングがいずれも有意に減少し、GlcNAc6ST-1が合成するL-セレクチンリガンドはリンパ球ホーミングに関与することが明らかとなった。ポリ-N-アセチルラクトサミンの分岐構造を合成するβ-1、6-N-アセチルグルコサミン転移酵素(IGnT)遺伝子のノックアウトマウスも作成した。このマウスは活動性が低下し、現在その原因を解析中である。また、リンパ球数も減少していたが、赤血球数は変化しなかった。
