サイトカインの生理活性発現における糖鎖認識機構の関与

研究概要	様々な液性免疫因子がレクチン様活性を有しており、複合糖質の糖鎖が生理活性のモジュレーターとして機能していることが明らかになりつつある。我々は多様なサイトカインの固有な糖鎖認識と生理活性発現との関連を明らかにすることを目的として研究をおこなっている。現在までのところ、サイトカインはその結合特異性からプロテオグリカンを認識するグループ、高マンノース型糖鎖を認識するグループおよびGPIアンカー糖鎖を認識するグループの3種に分類されるが、プロテオグリカンを認識するグループを除く2つのグループの存在は我々の研究から明らかになったものである。平成14年度はインターロイキン18(IL-18)がGPIアンカーグリカン部を認識するサイトカインであることを発見し、その結合特異性と生理活性への関連について以下の知見を得た。IL-18刺激によってIFN-γを産生するヒトKG-1細胞を用いてGPIアンカーグリカン部の部分糖鎖添加の影響を調べた。その結果、マンノース-6-リン酸を添加すると阻害作用がみられる一方で、マンノビオースジリン酸を加えるとIL-18効果の増強がみられ、同時にチロシンキナーゼ活性が数十倍に増強した。これらの現象に加えて[^<35>S]標識IL-18にマンノース-6-リン酸あるいはマンノビオースジリン酸を共存させるとそれぞれ単量体および二量体が主成分となった。したがって、GPIアンカー糖鎖上のマンノビオースジリン酸エステルを認識して形成された二量体IL-18がIL-18受容体との高親和性複合体形成を促進し、シグナル伝達を引き起こすものと考えられる。
