サイトカインの生理活性発現における糖鎖認識機構の関与

研究概要

様々な液性免疫因子が糖鎖認識活性を有し、複合糖質の糖鎖が生理活性のモジュレーターとして機能している。我々は平成14年度に続き、IL-1β、IL-18のみならず腫瘍壊死因子(TNF-α)がGPIアンカー糖鎖タンパク質に結合し、その結合がマンノース6リン酸で阻害されることを見出した。まず、GPIアンカーの糖鎖構造をヒト胎盤アルカリホスファターゼ(hAP)を材料にして詳細に調べた。その結果、GlcNAcβ1→リン酸がコア構造の2番目のマンノースのC-6位に結合していることが明らかとなった。hAPからGlcNAcβ1→リン酸を除去するとTNF-αはhAPに結合しなくなることから、このマンノース6リン酸ジエステルがTNF-αに結合すると考えられる。そこでTNF-α刺激によりアポトーシスが誘導されるマクロファージ系細胞株U937にマンノース6リン酸を添加するとアポトーシスが100%阻害されることが判明した。さらに、PI-PLCで処理した細胞およびマンノース6リン酸添加でチロシンのリン酸化も阻害された。したがって、TNF-αはそのタンパク質性受容体と結合すると同時にGPIアンカー糖鎖を認識し、高親和性の(TNF-α)_3(TNFR)_3(GPI)_3のオリゴマーを形成することによって、アポトーシスを誘導すると考えられる。一方、昨年度報告したIL-18ではTNF-αと異なって、GPIアンカーの2つのマンノース6リン酸ジエステルにそれぞれ1モルのIL-18が結合し、タイトな2量体を形成し、これが、IL-18Rα,βとの高親和性複合体形成のトリガーとなることを明らかにした。現在、ケモカインであるIL-8とIL-15がユニークな糖結合活性を示すことを見い出し、それぞれの生理機能活性をモジュレートしていることを追跡中である。
