塩類細胞の分子解剖と分化誘導機構

研究概要

水生生物の存在基盤となっている塩類細胞の機能と分化誘導機構を理解するために,本年度は淡水型と海水型塩類細胞の分子解剖及び分化誘導因子の候補分子の探索を行い以下の成果を得た。1.恐山ウグイの酸性適応機構の解明:pH3.5に適応させたときにエラの塩類細胞に強<発現してくる分子群の構造・機能解析から,重炭酸イオン輸送分子で体液の酸性化を防ぐ役割を果たしているNa^+/HCO_3^-共輸送体(NBC)がこれまで知られているものに比べイオン親和性が強いなどのユニークな性質を有することを明らかにすると共に,グルタミン代謝系を活性化し,アンモニアと重炭酸イオンを生成し酸性に適応しているという驚くべき仕組みを解明した。2.淡水ウナギのエラ上皮細胞で特異的に発現するRBCCタンパク質の同定と局在部位の決定:転写調節因子様のドメイン構造を有するRING-finger B-box coiled coil (RBCC) タンパク質を同定し,エラでのみ特異的に発現し,淡水で発現量が著しく増加することを明らかにした。さらに抗体染色により塩類細胞を中心とするエラの上皮細胞の核内に多く存在することを見出した。3.淡水型塩類細胞の機能解析:過剰の塩を排出するという海水型塩類細胞の役割と分子機構の理解に比べ,淡水型塩類細胞の機能は分子レベルではほとんど分かっていない。淡水から塩を吸収するという難しい仕事を担っていると考えられているので,それらの輸送に関与する候補分子をクローニングし,機能解析の基盤を築いた。
