研究概要

下痢症ウイルスの診断法の開発として3つのことを行った。(1)昨年度のノロウイルスに対するイムノクロマト法の世界での最初の開発ではLordsdale近縁のウイルスを中心に検出できた。今年度は、すべてのノロウイルスあるいはサポウイルスに対して検出できるイムノクロマトを開発するためにバキュロウイルス系を用いて発現を試みた。現在発現を見出し、抗体の作成を行っている。(2)2つのmultiplex PCR法による下痢症ウイルスの検出系を完成させた。1つはロタウイルスA, B, C群とアデノウイルスを検出するセットであり、もう1つはノロウイルスGI, GIIとサポウイルス、アストロウイルスを検出するセットである。(3)環境との関連で、2年にわたり不忍池の水を集めている。ヒト、トリに関してPCR法による遺伝子検出を可能とした。
疫学調査として、わが国の5カ所の小児の下痢便中のウイルスを継続的に調べている。舞鶴でC群ロタウイルスの集団発生が見いだされたさらにA精神障害施設でロタウイルス、アストロウイルスの集団発生、あるいはB病院で職員と患者でノロウイルスの集団発生、そして個別的にはロタウイルス脳症の発症を調べた。平成14年-15年ではわが国においてはI型が減少し40%ぐらいとなった。また2型と9型が増えてきた。またノロウイルスではGIIが依然として主流をしめており、とくにLordsdale近縁のゲノタイプであった。環境中のウイルスの検出系を開発するために、陰電何膜を利用して濃縮する方法を用い、大学に近い不忍池の水で経年的に調べたが、ヒトの下痢症ウイルスを検出するまでになっていない。
食品、特に牡蠣の中のノロウイルスを全国調査で調べた。9%(17/191)の牡蠣にノロウイルスを見出した。GIが3個、GIIが14個であった。多くは10^2以上のコピー数があった。したがって生牡蠣は重要なノロウイルスの感染経路となりうる。また集団食中毒ではA型肝炎ウイルスとノロウイルスが同時に見られたことがあった。
可視光応答型光触媒酸化チタンの抗下痢症ウイルス作用について調べたところ、わずかながら作用が見られた。
