皮膚疾患におけるケモカインと樹状細胞の関与についての検討

研究概要	樹状細胞については、マウス表皮から純化して採取したランゲルハンス細胞(LC)を用いてLCによるTARC、MDCなどTh_2ケモカインとIP-10、Mig、IPACなどTh_1ケモカインの産生についてmRNA、蛋白レベルにおいて制御機構についても検討を加え、現在まとめている。またLCおよび脾樹状細胞(DC)のLPSに対する反応について検討すると共にToll-like Receptor(TLR)の発現と、それぞれのリガンドによる反応についても検討を加え、それぞれを現在まとめている。ケラチノサイト(KC)からのTARC、IP-10産生とその制御について検討し、報告した。IFN-γとTNF-αによって産生されるTARCとIP-10はIFN-γ+TNF-αにIL-4を加えることによってそれぞれ抑制、増強されることが明らかとなった。臨床的にはTh_2プロフィルを示す水疱性類天疱瘡と菌状息閉症においてKCからTARCが産生され水症液中、血清中TARC産生の上昇がみられ、病勢を反映することを示した。アトピー性皮膚炎では血清MDCが上昇し、病勢を反映することを示した。ヒト末梢血からの樹状細胞の誘導に成功し、それを用いた発現分子制御、サイトカイン産生制御について検討を進めている。
