血液脳関門機能障害と精神疾患に関する生体膜輸送生理学的研究

研究概要

抗てんかん薬のバルプロ酸が血液脳関門を血流律速の担体輸送系で排出されることを明らかにした。血液脳関門障害モデルとして、ジエチルマレイン酸処理による酸化的ストレス効果を解析したところ、グルタチオンの前駆体であるシスチン輸送担体system Xc^-が顕著に誘導されることを明らかにした。輸送活性の増加だけでなく、mRNAも顕著に誘導された。一方、血液脳関門障害モデルとして高浸透圧処理を行ったところ、神経伝達物質の一種であるプロリンの輸送系system Aの輸送活性が増加することが明らかになった。さらに、定量的PCR法を用いてsystem Aの候補遺伝子ATA1,ATA2,ATA3の誘導効果を解析したところ、ATA2が最も顕著に誘導されることが明らかになった。この輸送系の活性化は、脳内浸透圧調節と脳毛細血管内皮細胞の構造維持に重要な役割を果たしていると推察される。一方、脳神経保護効果が知られるbasic FGFの血液脳関門輸送機構を解析した。その結果、脳側細胞膜にbasic FGFを内在化する機構が存在することが示唆された。basic FGFは星状膠細胞で生成されることから、この輸送機構の知見は、脳毛細血管内皮細胞に対する栄養因子効果の解析において有用である。脳内エネルギー蓄積物質としてクレアチンは非常に重要な役割を果たしている。血液中のクレアチンは血液脳関門のクレアチン輸送担体によって、脳内へ供給されることが明らかになった。神経保護効果が注目されている脳内クレアチンの主要供給経路が明らかになったことから、クレアチン代謝異常が関係する精神神経疾患の治療に有用な示唆を与えるものである。
