"プロトンブロック法"によるDNA・RNAの究極新合成法の開拓

研究概要	本研究は、分子生物学や、医学・薬学の分野はもとより、核酸を分子工学的素材として活用する新世代の機能性核酸の研究の飛躍的な発展に不可欠な、従来の合成法を凌駕する画期的なDNA・RNAの化学合成法を開拓する基礎研究をおこなうことを目的に開始したものである。現在用いられているDNAやRNAの化学合成法は、単純な未修飾のDNAやRNAを合成するために開発されたものであり、tRNAやrRNAのように複雑な修飾塩基を含むRNAの合成には応用できないのが現状であったが、本研究ではこの現状を打開するために全く新しいコンセプトを提案し、実際に極めて将来有望と思われるDNAの化学合成法を2つ開拓することができた。ひとつはプロトンブロック法と呼ばれる合成法であり、もうひとつは活性ホスファイト法と名付けたものである。とくに、後者の方法はキャップ化反応も省きながらDNA20量体の合成を達成したものであり、これまでの常識を覆す画期的な研究成果である。ピリミジンオリゴヌクレオチドに関しては、さらに長鎖なものも合成可能と思われるので、本法はアンチジーン法への展開も期待される。プリンオリゴヌクレオチド合成に関しては、グアノシンの縮合効率の改善とアデニン塩基部位への副反応の回避がこれからの新しい課題となった。これらの問題点はグアノシンの場合には5'水酸基の保護基の脂溶化、後者の場合にはアルコール系活性化剤の開拓によって解決できる目処がたっている。今後は、この基礎研究をもとに数多くの応用を手がけ、この合成法を多種多様なハイスループット型遺伝子診断技術に効率よく活用していく予定である。
