研究概要

1.GAGA因子と複合体を形成するFACTが、ヌクレオソームを変形してクロマチンリモデリング因子の作用を促進するため、GAGA因子の結合配列近傍で特異的にリモデリングが起きることを明らかにした。
2.GAGA因子と相互作用しているタンパク質群としてFACTの外に、クロマチンリモデリング因子Brahma複合体のサブユニットを同定した。この結果は、GAGA因子-FACT複合体によって誘起されるクロマチンのリモデリングにBrahma複合体が関わることを示している。
3.遺伝学的解析により、生体内でGAGA因子-FACT複合体がHox遺伝子群のエピジェネティクな発現制御に関わることを示した。
4.遺伝学的および、生化学的解析により、GAGA因子-FACT複合体がposition effect variegationに関わることを明らかにした。
5.個体レベルでのRNAiを用いて超らせん化因子がX染色体の量的補正に関わることを発見した。
