ヘテロ多重結合での結合生成に基づく新しいラジカルカップリング反応

研究概要	炭素-炭素多重結合におけるラジカル的な炭素-炭素結合生成反応は極めて有効な炭素管格構築法であり,広く有機合成に用いられている。それに対し炭素-ヘテロ多重結合の利用は極めて限られている。本研究では、申請者らが最近見出したシリルテルリドとヘテロ多重結合との反応による炭素ラジカルの新しい発生法の発展をはかることで,炭素-ヘテロ多重結合における新しいラジカルカップリング反応の開発について検討を行った。シリルテルリドを用いたカルボニル化合物とアルキンとの熱的カップリング反応について検討を行ったところ、期待通りにビニルテルリド構造を持つグループ移動型のカップリング生成物が収率良く得られた。生成物のアルケン部位は高度にE-体に制御されており、多くの基質の組み合わせにおいて、ほぼ単一の生成物が得られた。さらに、生成物における炭素-テルル結合のラジカル的開裂反応を用いてアルケンと立体選択的に炭素-炭素結合生成が行えることを明らかにした。このことから、本反応がシリルテルリドとスズヒドリドとを、順次用いたカルボニル化合物、アルキン、アルケンとの逐次カップリング反応による三置換アリルアルコール類の立体選択的合成法となることを明らかにした。カップリングの基質としてカルボニル化合物の代わりにイミンを用いることができると、アミンの合成法と成り得る。そこで、種々のシリルテルリドとイミン、イソニトリルとのカップリング反応について検討を行った。その結果、新しくトリエチルシリルシリルテルリドを開発することで、望むカップリング反応が効率的に進行することを明らかにした。さらに、生成物の炭素-テルル結合の変換反応を用いることで、種々のアミノ酸誘導体のコンビナトリアル合成法として優れていることを明らかにした。
