卵成熟におけるサイクリンB翻訳開始機構の分子形態学的解析

研究概要	卵成熟における翻訳開始とカップリングしたサイクリンBmRNAの形態変化に関わる分子同定と機能解析、及び、これら分子とすでに翻訳調節に関与することが示されているCPEB並びにPumilioとの機能相関を軸に研究した。サイクリンBmRNA凝集体形成に関わる蛋白質同定に関しては、ビオチン化mRNAを用いてmRNA結合蛋白質のアフィニティー精製を行い、いくつかの特異的なバンド(32、33、48、49、51、52 kDa)の検出に成功した。これらを切り出し、N末アミノ酸配列の分析を行ったが、いづれも、N末ブロックあるいは蛋白質量の不足のため、配列決定に至らなかった。N末がブロックされているものについては、トリプシン分解後、HPLCによりペプチド断片を回収し、アミノ酸配列を決定しつつある。また、量的に少ない蛋白質にういては、スタート材料を増加し、配列決定に必要な資料を得た。 CPEB並びにPumilioとサイクリンBmRNA翻訳開始に関しては、これらの組換え蛋白質や抗体を用いた実験を行い、1)サイクリンB1の3'非翻訳領域に存在するPumilio認識配列に変異を持つレポーターは、野生型のそれより効率よく翻訳されること、2)Pumilio抗体を注射された卵母細胞ではホルモン刺激後の卵成熟が加速されること、3)Pumilio RNA結合領域を過剰発現した卵母細胞では、ホルモン刺激後のサイクリンB1 mRNAのポリアデニル化の阻害を介した翻訳阻害により卵成熟が遅れること、4)PumilioとCPEBは未成熟卵内で結合しているが、成熟過程でリン酸化CPEBとPumilioとの結合が消失することが明らかになった。これらの結果は、PumilioはサイクリンB1mRNAのポリアデニル化の阻害を介して翻訳を抑制する機能を持ち、その抑制解除にリン酸化CPEBとの解離が関わることを示唆する。
