イネにおける個葉の光合成能力の改善と個体バイオマスの向上をめざして

研究概要	光合成炭酸固定酵素Rubiscoの小サブユニット遺伝子rbcSのpromoterとcDNAをアグロバクテリウムの形質転換用ベクターpBI101Hmにセンス方向とアンチセンス方向に組み込み、Sense-rbcSイネとAnti-rbcSイネを作出した。Anti-rbcSイネ作成においては、極端にRubisco量の少ない個体を得るために、再分化から選抜そして育成の過程をすべて高CO_2グロースチャンバー(100PaCO_2)内で行った。形質転換体の導入遺伝子とコピー数の確認を行い、一部の個体についてはガス交換法による光合成測定も行なった。 Sense-rbcSイネにおいては、Rubisco量比が120%を越えるものが4個体得られ、Anti-rbcSイネでは、Rubisco量比が40%未満のものが2個体得られた。選抜4個体のSense-rbcSイネはすべて稔性を持ち、種子を得ることができた。当代個体は株保存後、再栽培を行いガス交換による光合成解析を行ったが、現在の段階ではRubisco-limitの領域(葉内CO_2分圧20Pa)においても光合成速度の増加は認められなかった。また、再栽培個体ではRubisco量の増加率も0〜15%程度まで低下していた。その原因について、rbcSとrbcL mRNAの発現量へのsense効果を含めて現在検討中である。また、Sense-rbcSイネの新規作成も継続して行っている。Anti-rbcSイネにおいては、クロロフィル蛍光測定の結果、常に高いNPQ(non-photochemicalquenching)が観察され、キサントフィルサイクル色素のデエポキシ化が進んでいることが認められた。P700測定の結果、このNPQ形成には、通常のイネでは限られた条件でしか観察されないPSIでの循環的電子伝達が働いていることが示唆された。
