イネにおける個葉の光合成能力の改善と個体バイオマスの向上をめざして

研究概要

光合成炭酸固定酵素Rubiscoの小サブユニット遺伝子rbcSをセンス方向に組み込んだ形質転換体Sense-rbcSイネのT_0世代において、Rubisco量比が非形質転換体(wild-typeイネ)に比べ120%を越えるものが4個体得られた。それらの効果に対して、sense遺伝子のコピー数との関係は明確でなかったが、antisense遺伝子に関してはある程度の相関(コピー数1〜5)が観察された。mRNAとの比較においては、sense個体においてrbcS mRNAの増加は認められたもののrbcL mRNAには明確な増加は認められなかった。コピー数2、Rubisco量121%のSense-rbcSイネ#35の次世代(T_1)の20個体について葉位を追って、第6葉身、11葉身、13葉身(止葉)のRubisco量を調べたところ、wild-typeイネのRubisco量に対し195±2%という大きなRubisco量増加が、しかも、葉位を問わず安定的に認められた(#35-8)。トウモロコシ葉における葉肉葉緑体と維管束鞘画分に分配された光合成構成成分比を調べた。電子伝達の構成成分であるCyt f量比は葉肉葉緑体に58%、維管束鞘画分に42%で、CF_1はそれぞれ44%と56%であった。両画分に葉緑体に局在するNADP-G3P-DH活性の葉肉葉緑体への活性分配比から、全Cyt f量のうちのRuBP再生産系分配相当量を算出した。その結果、68%相当量のCyt fがRuBP分配相当量と見積もることができた。しかし、葉身窒素含量当たりのCyt f量はトウモロコシにおいてイネの1.5倍程度であったので、トウモロコシにおいて窒素含量あたりのRuBP再生産系Cyt f量が必ずしも多いわけではないことが推定された。
