動物におけるアフラトキシン解毒機構の研究

研究概要

ラット・マウス・ハムスター・ブタ・ニワトリ・マスの肝臓サイトゾール分画について、ハムスター肝臓ミクロゾーム、補酵素、AFB1(アフラトキシンB1)、GSH(グルタチオン)の存在下で反応させ、生成されたAFB1-GSHをHPLCで定量することによってAFB1に対するGST(グルタチオン S トランスフェラーゼ)活性を求め、動物種間での比較を行った。生成されたAFB1-GSH量は、反応液よりSepPakC18で精製したAFB1-GSH分画を、HPLC (aminopropyl silicaに共有結合したD-phenylglycineで充填剤したchiral column)に供し測定した。そのデータから各動物種におけるGSTのKmおよびVm値を明らかにした。また、AFB1-aldehyde reductase活性測定のために、基質であるAFB1-aldehydeと反応生成物であるAFB1-dialcoholeを化学的に合成し、各種動物種肝臓サイトゾールによるAFB1-aldehydeからのAFB1-dialcohole生成をHPLCにより測定することによって各種動物のreductase活性を明らかにした。
