脂肪酸結合タンパクの機能解析、特に神経系と免疫系に注目して

研究概要	我々が世界に先駆けて作成したE-FABP(皮膚型)遺伝子欠損マウスの表現型として、1)表皮におけるH-FABP(皮膚型)の遺伝子発現の亢進、2)正常時における表皮を経由する水分放散量が野生型に比べて減少すること、および有機溶剤処理後の水分放散防御能の低下後の正常時状態までの回復が野生型における経過に比べて著しく遅延すること、が認められた。後者はヒトでは老人皮膚の状況に類似していた。皮膚以外ではE-FABPは、胸腺皮質の上皮細胞で特異的に発現されていることが初めて見出された。この所見は昨年発見して公表した肺胞マクロファージュにおけるE-FABPの強発現と併せて、このタンパクの抗原提示機能への深い関与を強く示唆している。さらに、免疫組織化学的にこの種の細胞の全貌を明瞭に顕微鏡下で観察可能にするためにはE-FABPが有用なマーカーとなることが判明した。同じく世界に先駆けて作成したB-FABP(脳型)遺伝子欠損マウスの表現型としては、1)B-FABPがアストログリアに発現局在するにも拘らずニューロンでE-FABPの遺伝子発現の軽度亢進を示すこと、2)B-FABPに好んで結合するドコサヘキサエノイン酸(DHA)の脳内含量が有意に低下すること、および3)行動学的解析により学習と記憶が低下していること、が明瞭に認められた。DHAの極低含量餌飼育による記憶低下の報告はこれまでにあるが、DHAの細胞内局在に直接関与するFABPの発現有無により、個体の学習記憶が影響されることを明示した今回の所見は、DHAの機能解明に大きな寄与をなすものと考えられる。これらの所見を下に、15年度は両FABPの生体での機能解明に神経と免疫双方から努力する予定である。
