研究概要

今年度はRestriction Landmark Genomic Scanning (RLGS)法の予備実験から研究を開始した。検討の結果、生殖器系の精巣上体、精巣、子宮では2〜3匹分のゲノムDNAからRLGS法によるメチル化解析が可能であった。
臓器間のDNAメチル化の違いを精巣上体と子宮で比較したところ、ゲノム上の約1000ケ所のうち、2ケ所(0.2%)が精巣上体のみで脱メチル化されており、1ケ所(0.1%)が子宮のみで脱メチル化されていた。合わせて3ケ所(0.3%)のメチル化が異なっていたが、これは臓器間のDNAメチル化を比較した過去の報告と比較しても妥当な数であると思われた。
内分泌撹乱物質として知られる合成エストロゲンのジエチルスチルベストロール(DES)をC57BL/6マウスの新生仔期(生後1〜5日)にDES3、0.3、0.03、0.003μg/dayを投与し、30日齢で精巣上体、子宮からゲノムDNAを抽出し、ゲノム上の約1000ケ所の領域についてRLGS法によりDNAメチル化の状態を解析した。
その結果、DES3μg/dayの新生仔期投与により、精巣上体で6ケ所の脱メチル化と1ケ所のメチル化、子宮で6ケ所の脱メチル化と5ケ所のメチル化が検出された。DES投与後の精巣上体で脱メチル化された6ケ所中の1つは、子宮ではいずれの場合も脱メチル化されている部位であり、メチル化された1ヶ所はいずれの子宮でもメチル化されている部位であった。このことから、DES投与後の精巣上体のDNAメチル化は子宮に似たパターンを示すことが示唆された。
来年度は今年度の成果を発展させ、DES投与によりメチル化が変化したゲノム領域のクローニングや、DES投与濃度によるメチル化の変化と形態的変化の関係等を調べる予定である。
