中枢神経系発達における甲状腺ホルモンの作用機序とホルモン作用臨界期の形成機構

研究概要	甲状腺ホルモン(thyroid hormone, TH)は中枢神経系の正常な発達のために不可欠である。THは特定の臨界期のみ中枢神経系に作用し、発達を調節する。本研究はげっ歯類小脳を用いて中枢神経系発達に及ぼすTHの作用を解明する事を目的として行われている。発達期小脳において、THは臨界期(げっ歯類は生後約2週間)のみに標的遺伝子発現を調節する。しかしTH感受性変化の機構は明らかではない。TH受容体(TR)発現は発達につれ増加するので、感受性低下には別の機構が関与する可能性が高い。そこで我々は、中枢神経系で強く発現するTRの転写共役因子のSRC-1の関与をまず調べた。発達期小脳におけるSRC-1の発現を免疫組織化学的に調べた結果、プルキンエ細胞に強い発現が見られた。また、内顆粒層にも生後2日めから発現している事が分かった。しかし、外顆粒層には発現が見られなかった。この事から、TRの作用に関して細胞特異性がある可能性が示唆された。同時にyeast two hybrid systemを用いて発達期小脳におけるTR結合蛋白のスクリーニングを開始した。そして、数個のpositive cloneが採取された。次年度はこのcloneを更に詳細に解析する予定である。また、臨界期に環境ホルモン等によりTHの作用が撹乱された場合、クレチン症類似の影響が出る事が知られている。そこで、transient transfection法を用いてTRによる転写調節への内分泌撹乱物質の作用を調べた。その結果、10-10Mという低用量のPCBがTRを介する転写を抑制した。抑制作用は線維芽細胞よりも小脳顆粒細胞由来の株細胞で強かった。この事は脳発達の臨界期に内分泌撹乱物質に暴露した場合、低濃度でもTRによる転写機能に影響が出現する可能性を示唆している。現在、メカニズムの更なる解析を進めている。
